Kisspeptin

A neuropeptide that acts as the master upstream regulator of the reproductive hormone axis, stimulating GnRH, LH, and FSH release.

Kisspeptin is a naturally occurring neuropeptide encoded by the KISS1 gene that serves as the primary gatekeeper of puberty onset and reproductive function. It stimulates gonadotropin-releasing hormone (GnRH) neurons in the hypothalamus, triggering the downstream release of LH and FSH. Clinical studies have demonstrated its ability to restore LH pulsatility in hypothalamic amenorrhea and induce egg maturation in IVF patients.

⚙️ Mechanism of Action

Kisspeptin binds to the GPR54 (KISS1R) receptor on GnRH neurons in the hypothalamus, directly stimulating GnRH secretion into the hypophyseal portal system. This triggers pulsatile release of LH and FSH from the anterior pituitary, activating gonadal steroidogenesis. Unlike exogenous GnRH analogs, kisspeptin works upstream of the HPG axis and maintains physiological pulsatility rather than causing receptor desensitization.
