PT-141 (Bremelanotide)

Melanocortin agonist for sexual function

PT-141 (Bremelanotide) is a synthetic peptide analog of alpha-melanocyte-stimulating hormone. Unlike PDE5 inhibitors, it works through the central nervous system to enhance sexual desire and arousal. FDA-approved as Vyleesi for female hypoactive sexual desire disorder.

⚙️ Mechanism of Action

PT-141 activates melanocortin-4 receptors (MC4R) in the hypothalamus, directly stimulating sexual arousal pathways in the brain rather than acting on the vascular system.

Benefits

  • Enhanced sexual desire and arousal
  • Works via central nervous system (not vascular)
  • Effective in both men and women
  • FDA-approved (Vyleesi) for HSDD

⚠️ Risks & Considerations

  • Nausea (most common side effect)
  • Facial flushing and headache
  • May increase blood pressure transiently
  • Not for use with certain cardiovascular conditions
