A mitochondrial-derived peptide that acts as an exercise-induced metabolic regulator, improving insulin sensitivity and physical performance.
MOTS-c (mitochondrial open reading frame of the 12S rRNA-c) is a 16-amino-acid peptide encoded within the mitochondrial genome that functions as a signaling molecule between mitochondria and the nucleus. Exercise increases endogenous MOTS-c levels up to 12-fold in skeletal muscle and 1.5-fold in circulation. It promotes metabolic homeostasis, improves insulin sensitivity, and has been shown to reverse age-related physical decline in animal models.
⚙️ Mechanism of Action
MOTS-c acts primarily through the Folate-AICAR-AMPK pathway, a central metabolic sensing axis that regulates energy balance. In response to metabolic stress, MOTS-c translocates from the cytoplasm to the nucleus where it regulates adaptive gene expression programs. It activates AMPK signaling to enhance glucose uptake and fatty acid oxidation, mimicking many of the metabolic benefits of physical exercise at the cellular level.
